​​​​​​​The increasing penetration of distributed energy resources (DERs), including photovoltaic systems, battery energy storage systems, HVAC systems, and flexible loads, is transforming conventional power systems into decentralized, bidirectional, and market-interactive networks. This dissertation develops a transactive energy framework for coordinating distribution system operators, aggregators, and prosumers while maintaining distribution network reliability. The work first examines the evolution from peer-to-peer energy trading toward broader transactive energy structures, emphasizing the need for market mechanisms that are compatible with physical grid constraints. A hierarchical DSO-aggregator coordination architecture is then proposed in which the DSO certifies network-aware export limits by evaluating voltage and thermal constraints, while aggregators manage DER portfolios to provide grid services. The framework is extended across multiple timescales by coupling day-ahead and intra-day market clearing with model predictive control (MPC)-based DER dispatch. Simulation studies using modified IEEE distribution test systems demonstrate that the proposed MPC-based approach enables cost-effective aggregator selection, adaptive redispatch, and reliable tracking of market commitments while keeping aggregator operation within network-certified limits. The results show that coordinated transactive energy markets can support DER participation, improve operational flexibility, and provide a practical foundation for future distribution-level market architectures.

Novice learners in introductory computer science (CS1) courses frequently face steep cognitive hurdles when grappling with invisible system behaviors, execution mechanics, and syntax-dense abstractions. Among the pedagogical approaches used by cs educators, Notional Machine is a signature pedagogical device that supports novice learners’ conceptual development by simplifying abstract concepts using analogies, metaphors, and various forms of simplified representations. While educators have historically employed text- or screen-dependent “notional machines” to form mental models, traditional approaches are often passive, teacher-centric, and exclude the diverse cognitive needs of learners. This thesis addresses the systemic barriers and limitations of traditional NMs by introducing an accessibility-first approach to design and develop notional machines that provide both “low floors” for immediate access and “ramps” for inclusive interaction. The goal of this thesis is to achieve the curb-cut effect by incorporating design patterns of assistive computation learning interventions into notional machines and demonstrate the design, development, and impact of notional machines for assistive learning (NMAL) through collaboration with CS educators and students. This dissertation demonstrates that notional machines designed with an accessibility first lens minimize systemic execution gaps and catalyze equitable conceptual growth. The resulting artifact blueprints, co-design methodologies, and scaling guidelines form a transformative framework for making computer science education universally accessible

The increasing penetration of inverter-based resources (IBRs) is leading to a significant reduction in the effective inertia of modern power systems. Moreover, the intermittent and stochastic behavior of IBRs results in time varying system inertia, posing critical challenges for frequency stability and system reliability. This dissertation introduces a novel datadriven inertia estimation framework for both large disturbance events and ambient operating conditions. This capability enables not only event-based inertia assessment following major disturbances, but also continuous monitoring of system inertia during normal operating conditions. The proposed framework is capa ble of estimating both synchronous inertia and virtual inertia contributions in unit level as well as systemwide. Generator outages induce large disturbances, while ambient conditions are modeled through dynamic load changes. The estimation process begins with Detrended Fluctuation Analysis (DFA) to accurately detect the onset of a disturbance, after which a modified autoregressive moving average exogenous input (MARMAX) model is employed to estimate each generator’s inertia constant and a polynomial curve fitting approach for system level inertia estimation. The primary contributions include the application of the estimation framework under diverse system operating condition i.e. ambient or disturbances, precise event detection via DFA, and optimizing window selection for accurate inertia estimation. Validation on the IEEE 39bus transmission system under generator outage and dynamic load conditions, along with tests on real event data from PMU in the US Eastern Interconnection, demonstrates that the proposed framework significantly enhances accuracy and efficiency compared to existing methods."

The increasing penetration of distributed energy resources (DERs), advanced power electronics interfaces, and dynamically reconfigurable switching operations has transformed modern electric distribution systems into highly nonlinear, unbalanced, and data-rich networks. While these advancements improve grid flexibility, they also introduce significant challenges for real-time monitoring, accurate modeling, and reliable voltage regulation. Conventional analysis tools are highly dependent on fixed system parameters, static topology information, and balanced approximations that are no longer valid under high DER variability, frequent switching, and unbalanced loading conditions. As a result, utilities face limited observability, inaccurate state estimation, and reduced effectiveness of traditional voltage control devices such as capacitor banks and on-load tap changers (OLTCs). Addressing these challenges requires a unified framework capable of continuously estimating system parameters, identifying the actual operating topology, and coordinating DER-based voltage support with legacy devices in real time.

This dissertation proposes an integrated methodology for three-phase unbalanced distribution systems that brings together: a hybrid Kalman filter estimation algorithm for real-time estimation of line conductance (G) and susceptance (B), leveraging a physics-informed path matrix to ensure numerical stability, phase-selective estimation, and fast convergence under changes in load and switching conditions; a topology estimation framework based on Thevenin impedance signatures at switch locations, capable of identifying open/closed statuses across multiple feasible network configurations without requiring exhaustive search or full observability; and a data-driven voltage control algorithm that optimally coordinates DER reactive power injections with legacy devices, ensuring rapid voltage recovery following disturbances and minimizing tap operations.

Overall, this dissertation provide design and development method for a unified framework that enhances distribution systems situational-awareness, improves parameter and topology accuracy in real time, and enable fast, reliable voltage regulation in the presence of high DER penetration, The outcomes support the development of next generation distribution management systems (DMS), providing utilities with scalable, data-driven tools for operating modern unbalanced distribution networks.

Abstract:

The altered gait patterns observed in individuals with chronic ankle instability (CAI) are associated with repetitive ankle sprains, episodes of giving-way, and altered loading patterns in the talar cartilage; thus, precipitating early onset ankle osteoarthritis. Laboratory/clinic-based gait interventions can alter these gait patterns and improve self-reported function among participants with CAI. To increase participant access beyond the lab/clinic, we aim to broaden the scope of gait interventions by leveraging individuals’ daily walking routines. Prior to developing such interventions, determining the biomechanical profile and tissue adaptations during walking in a real-world setting is necessary among individuals with and without CAI. Using a case-control design, 33 individuals with CAI and 33 healthy individuals walked a dedicated outdoor course while wearing accelerometers (RunscribeTM) tracking their biomechanical gait profile. Images of the talar cartilage were taken before and after the gait assessment to assess tissue response to loading in the real-world setting. Differences in biomechanical variables were assessed between groups using statistical parametric mapping, while repeated measures ANOVA were used to determine pre-to-post changes in cartilage outcomes. We found that individuals with CAI displayed altered biomechanics and different responses in talar cartilage outcomes when compared to healthy individuals.

Abstract:

Fluorescent small molecule voltage sensitive dyes (VSDs) are versatile and powerful reporters of the membrane potential across living cell membranes. They have made the mapping of coordinated activities of neurons and cardiac cells simple and efficient, which is essential for the detection of a myriad of neurological and cardiac abnormalities and diseases. Several VSDs already exist for understanding cellular physiology including intracellular communication via cell membrane voltage sensing; however, they have their drawbacks in terms of fluorescence intensity, response times and/or voltage sensitivity. Thiazolo[5,4-d]thiazole (TTz) dyes are a relatively new and promising class of VSDs. While TTz dyes have high photostability and extremely low cytotoxicity, the mechanistic basis of their voltage-sensitivity has never been explored. To address this, a new generation of asymmetrically substituted TTz dyes (asym-TTz) have been accessed, that are push-pull in nature, with a donor-pi-acceptor molecular structure. The asym-TTz dyes consist of one electron donating (dibutylaminophenyl) and one electron withdrawing side chain (carboxyphenyl) on either side of a rigid, planar, electron deficient pi-conjugated aromatic TTz core. These TTzs have been synthesized following a mixed-pot reaction, with the starting materials comprising of the aldehydes of the desired donor and acceptor groups, and dithiooxamide/rubeanic acid, much like the previous generation of asym-TTz VSDs. The new charge-transfer asym-TTzs show extremely high quantum yields in non-polar environments (~98% in hexane), brilliant cell membrane localization and voltage sensitivity of ~10% F/F per 100 mV, which is comparable to several other existing VSDs. In addition to that, these asym-TTz derivatives exhibit strong solvatofluorochromism with large Stokes shifts (>100 nm in polar solvents) and transition dipole moments higher than any other reported push-pull dye (>20 D). By computationally analyzing their behavior under an electric field, a surprising twist on either side of the TTz bridging unit in the excited state was observed in the asym-TTz molecule with carboxyphenyl acceptor group at more positive membrane potentials (depolarization). This correspondingly resulted in the decrease of the dye’s fluorescence intensity experimentally. In contrast, another very similar asym-TTz dye, with a carboxypyrazinyl acceptor group instead of carboxyphenyl group, showed no computational twist around the TTz core and remained largely insensitive to voltage when tested in cells. This novel mechanism, involving the dual twisting at the excited state of the molecule leading to its voltage-sensing behavior, can be leveraged to create newer classes of highly sensitive voltage reporter dyes for diverse sensing applications. Additionally, since these asym-TTz dyes produced such promising results, a library of asym-TTz dyes were synthesized for the purpose of gaining a deeper insight into the synthetic and photophysical characteristics of push-pull asym-TTzs. This comprised of changing the electron acceptor group of the asym-TTz molecules while keeping the electron donor group (dibutylaminophenyl) constant. The side groups explored included ketone, fluorophenyl methyl ester isomers, pyridyl methyl ester isomers, an alkene bond preceding the methyl ester, a thiophene methyl ester, and a nitrile group. It was revealed that while some molecules showed identical optical behavior, the reason behind that for each molecule varied depending on the electronic properties of the side groups, and their ability to perturb the intramolecular charge transfer (ICT). Some isomers showed drastically different emission profiles for the same reasons. This study enables us to develop a deep understanding of asym-TTz fluorophore behaviour at a very fundamental level, which is crucial for making strides in the improvement of VSD production and efficiency going forward.

Abstract:

Advances in Next Generation Sequencing (NGS) have dramatically increased the volume of biological data available for analysis, creating significant computational challenges, particularly for microbiome and virome research. Existing software often requires substantial memory resources, scales poorly on computing clusters, or lacks sufficient database coverage to fully characterize biological diversity.

To address these limitations, this dissertation presents three computational frameworks: MetaCerberus, MerCat2, and EpiVirQuant. MetaCerberus is a scalable protein annotation pipeline that uses Hidden Markov Models (HMMs) to provide efficient, low-memory functional annotation across the tree of life. MerCat2 is a database-independent k-mer analysis tool for nucleotide and amino acid sequences that incorporates novel algorithms to reduce memory requirements while enabling scalable distributed computing for large-scale comparative analyses. EpiVirQuant is an automated image-analysis pipeline that applies blind deconvolution techniques to enumerate and size Virus Like Particles (VLPs).

Together, these tools address critical bottlenecks in the analysis of large scale sequencing and imaging datasets by improving scalability, reducing computational resource requirements, and expanding analytical capabilities. Abstract:

Individuals with disabilities constitute the largest global minority group, accounting for 16% of the world’s population, of which 80% are acquired between ages 18 and 64 (World Health Organization, 2023). Hays and colleagues (1996) acknowledged disability as a distinct dimension of identity, differentiating between the impact of acquired and congenital disability on an individual's life. Identity reconstruction is often a challenging process for those with acquired disabilities, especially for racial minorities, who must simultaneously navigate the dual marginalization of their racial and disability status, as well as its interaction with other identities they might hold. The study explores the lived experience of disability identity development in individuals with acquired disabilities from historically marginalized racial groups using an interpretative phenomenological analysis (IPA) framework. The study involved eight participants who self-identified as BIPOC individuals with acquired physical disabilities. The findings reflected the impact of intersectional marginalization on racial minorities with acquired disabilities, who grapple with racial and disability-related discrimination, stigma, barriers to education and employment and a healthcare system ill-equipped to support them. The findings offer important implications for clinical training and practice for medical and mental health professionals, such as the field counselor education and disability-related public health policy.

Abstract:

Individuals engage in careful consideration and preparation prior to dissent because conversations of upward dissent pose significant social and material risks (Bisel & Arterburn, 2012; Kassing, 1997; Rothschild & Miethe, 1996; Stewart, 1980). During preparation, individuals rehearse the upcoming interaction (Bisel et al., 2025) to practice managing impressions with important others (Schlenker, 1980). While typically users might prepare by imagining possible scenarios or consulting others, advancements in Generative artificial intelligence (GenAI) offer users a unique setting for rehearsal of important interactions by providing a private conversation and thought partner (Nilson, 2010). This dissertation aimed to examine the content and outcomes of users’ interactions with GenAI as they attempted to prepare for upward dissent. I used a sample of maximum variation and a mixed-method approach to examine data from open and closed-ended survey questions and GenAI interaction transcripts. I used this data to explore what individuals do during preparatory GenAI interactions and how they view the utility of that interaction. The findings are presented in four chapters. The first findings chapter highlights how preparation for upward dissent is accomplished through the co-construction of interaction by both users and the tool. I argue the interaction is constructed in three non-linear(iterative?) phases: detailing the issue, specifying the output, and refining the output. The second findings chapter provides a rich description of users’ experiences during the interaction that contribute to their experiences of rehearsal and catharsis during dissent preparation. The third findings chapter provides evidence for these types of GenAI interactions increasing users’ likelihood of dissent while reducing their communication apprehension around conversations about dissent. Finally, the last findings chapter examines the genre of GenAI responses and how it constructs the nature of dissenter labor through managerialist discourse. This dissertation concludes with a discussion and implications section, which highlights that GenAI use to prepare for upward dissent can be both a useful tool to prepare and a hegemonic device that invites dissenter labor while minimizing their agency in decision-making.
